Gram staining is a differential staining technique used to classify bacteria into Gram-positive and Gram-negative based on their cell wall structure. ⸻ 🧫 Principle • Gram-positive bacteria have thick peptidoglycan → retain crystal violet → appear purple • Gram-negative bacteria have thin peptidoglycan + outer membrane → lose crystal violet → take counterstain → appear pink/red ⸻ 🔬 Materials • Clean glass slide • Bacterial smear • Crystal violet • Gram’s iodine • Decolorizer (acetone/alcohol) • Safranin (or dilute carbol fuchsin) • Distilled water • Microscope + oil immersion lens ⸻ 📝 Procedure 1️⃣ Prepare smear → Air dry 2️⃣ Heat fix gently 3️⃣ Crystal violet — flood for 1 minute → rinse 4️⃣ Gram’s iodine — flood for 1 minute → rinse 5️⃣ Decolorizer — add for 10–20 seconds (until runoff clear) → rinse immediately 6️⃣ Safranin — counterstain for 30–60 seconds → rinse 7️⃣ Air dry → examine under 100× oil immersion ⸻ 🎯 Results • Gram-positive → Purple/Blue • Gram-negative → Pink/Red #veterinarymedicine #instagram #reelsofinstagram #reel #microbiology

🔬 Negative Staining Technique Negative staining is a simple staining method used to observe the true size, shape, and arrangement of microorganisms without heat fixing.
