The ability to program material structures with predictable material behavior such as hydrophilicity, degradation, swelling and diffusion opens the possibility to design the interface between material and living systems. The Hybrid Living Material (HLM) fabrication platform is an example of this as it combines programmable materials and programmable organisms mediated through computational design to achieve replicable fabrication of hybrid living materials. Hybrid living materials are characterized by the integration of engineered biological functionalities and precisely structured synthetic materials, resulting in dynamic and responsive behaviors such as sensing, actuation, controlled biochemical signaling, self-regulation, and environmental adaptability. The HLM fabrication approach uses multi-material 3D printed objects with controlled distribution of chemical signaling resins and subsequent attachment of hydrogel with engineered E. coli to construct biohybrid objects. The objects can signal to the E. coli and as a result, the objects demonstrate predictable, repeatable spatial control of protein expression. Computational design tools are used throughout the fabrication platform โ€” from the digital specification of chemical distributions to the simulation of the interaction between 3D printed objects and engineered living organisms. They enable to tune the response of the organisms themselves, adapt the object and its signaling properties or inform potential bioengineering work.
