Lowkey getting my PhD in making cells look pretty πŸ’… I’m currently working with RNA that encodes for a Green Fluorescent Protein (GFP), which is a protein that emits a green light when excited with a laser. The image of green cells is from an experiment where I transfected cells with GFP RNA. I love seeing the bright green color under the microscope and wanted to share it with you guys. But with a ~twist~ hehe There are lots of cell-based assays out there, including fluorescent cell staining (fire), cell counting with trypan blue (ice), methylene blue staining (water), MTT cell metabolism assay (earth), and bioluminescence assays (lightning). Seeing the invisible within cells πŸ”¬ Learning how intracellular signaling occurs inside cells can help scientists to better understand cellular behavior. But it can be difficult to observe these signals due to the small size of cells. Researchers introduced a technique to create multinucleated giant cells from the social amoeba Dictyostelium discoideum. The team used fluorescent probes to investigate cAMP and Ca2+ signaling within these enlarged cells. Live-cell imaging revealed that intracellular signaling is characterized by self-organized propagation events that align with cellular polarity – in a front-to-rear propagation pattern. This giant cell model may offer a promising strategy for dissecting the intricate spatiotemporal aspects of single-cell signaling. This research was published in Communications Biology. Yusuke V. Morimoto explains more in the Protocols & Methods and Cell & Molecular Biology Communities. This animation shows how dense clusters of vesicles may contribute to the rapid local clearance or buffering of cAMP, shaping intracellular gradients that support persistent polarity.
