Mitosis (normal cell division) as part of the cell cycle. Mitotic stages (interphase, prophase, metaphase, anaphase, telophase and cytokinesis) in onion root tip cells. DNA replication occurs at interphase during the so-called S phase (S = synthesis). This stage is followed by the G2 phase (G=gap) during which structures required for division begin to assemble. At interphase the chromosome material is dispersed within the nucleus and the nuclear membrane (envelop) is intact. Mitosis is announced in plant cells by the appearance of a circular pre-prophase band that consists of microtubules (not shown). A flat sheet of cytoplasmic strands (called phragmosome = partition body) is formed in the center of vacuolate cells, which helps the nucleus to migrate to a central position. Mitosis starts with prophase, a process during which the nucleus looses its membrane (nuclear envelop) and chromosomes spiralise. As chromosomes become shorter and thicker they appear first like threads (mitos = thread) and later like distinct rods. During interphase, the cell prepares for division. In prophase, the chromatin condenses into visible chromosomes. Metaphase is marked by the alignment of chromosomes along the cell's equator. Anaphase involves the separation of sister chromatids towards opposite poles. Finally, telophase concludes with the formation of two daughter nuclei. To prepare a slide to view onion root tip cells under a microscope, follow these steps: 1. Obtain a small section of an onion bulb. 2. Using a sharp razor blade or scalpel, carefully slice off a thin piece of the onion root tip. 3. Place the thin slice of onion root tip onto a clean microscope slide. 4. Add a drop of water or a staining solution (like iodine) to the onion root tip slice to enhance visibility of the cells. 5. Gently place a coverslip over the onion root tip slice to flatten it and prevent it from drying out. 6. Wipe away any excess water or staining solution from the edges of the coverslip. 7. Mitosis is the process by which eukaryotic cells duplicate themselves, forming two identical cells. This cell has been transfected with a fluorescent label to the histone H2B, which is concentrated in the cell's chromosomes. At this stage, the chromosomes have already been duplicated, and they are seen to split into two pairs, which are pulled to opposite sides of the cell. This is followed by a separation of the cell membranes (cytokinesis), leading to the formation of two identical daughter cells.
