The tumor microenvironment often suppresses the activity of T cells, hindering the effectiveness of cancer immunotherapies. A new study has explored a precise CRISPR-based gene-editing approach to overcome this challenge. By introducing naturally occurring dominant negative mutations in the FAS and TGFβR2 genes in primary human T cells using base editing, a team of researchers rendered them resistant to immunosuppressive signals in solid tumour environments. These CAR-T cells exhibited improved cytotoxicity, persistence, and reduced exhaustion markers compared to traditional overexpression approaches.
