CIRCADIAN ALIGNMENT AND METABOLIC CONTROL Research has tightly linked the intrinsic circadian timing system to human metabolism, with multiple independent lines of evidence pointing to circadian disruption as a driver of metabolic disease. First, in older men with insulin resistance, skeletal muscle shows disrupted 24-hour clock gene expression alongside a loss of rhythmic mitochondrial metabolism. Second, epidemiological studies and controlled laboratory shift-work simulations consistently demonstrate a strong association between circadian misalignment and metabolic dysfunction. Third, human pancreatic islets from individuals with type 2 diabetes exhibit reduced clock gene amplitude and impaired synchronizing capacity, while pharmacologically reinforcing the islet clock restores insulin secretion in diabetic tissue. Together, these findings indicate that disruption of circadian timing, and its alignment with the external environment, contributes directly to metabolic disease. The dominant circadian alignment signal is the natural light–dark cycle: bright, dynamic daylight during the day and darkness at night. Yet, modern Western societies spend ~90% of time indoors under low-intensity, static artificial lighting that lacks the temporal structure of natural daylight. Adding to this, recent human data show that simply working near a window, allowing the visual system to experience a rhythmic daylight signal, improves glucose homeostasis, increases whole-body fat oxidation, elevates evening melatonin, and modulates the human skeletal muscle clock, a key peripheral circadian oscillator. *This is a clip taken from my conversation with @dhrupurohit where we discussed all things light and human health.
