By sealing an upside-down container over a straw that leads to a lower exit point, the experimenter creates a system where gravity pulls the water down the "drain" tube. As this water falls, it creates a partial vacuum (low pressure) inside the upper chamber. The higher atmospheric pressure pushing down on the surface of the water in the open cup then forces that liquid up into the straw to fill the void.
