🧬Day in the Lab: measuring gene expression🧬 This RT/qPCR assay has been an absolute staple of my PhD so far! I use it all the time to measure the expression of certain genes in my adipocyte (fat cell) spheroids. At first I was looking at the genes we’d expect to see in an adipocyte so I could show that my spheroids are expressing the right thing! Now I’m able to use the same technique to see how my spheroids respond to different treatments. Prepare plasmids for DNA sequencing with me 🧬 A major part of my project is to study how different mutations to my protein of interest impacts its function. So, I make a lot of different mutations and combinations of mutations all the time. I also play around with different plasmid backbones for different types of experiments that I perform! When sending samples off for sequencing it’s important to know what DNA concentration to send the samples and if a sequencing primer is required or not. In this video, I prepared samples for “spot” checking meaning I only sequenced a portion of my plasmid. I could also sequence my whole plasmid when necessary! #phd #womeninstem #lab #research #science
