From plasmid to engineered cells 🧬🦠 Step 1 - Cell seeding HEK293T cells are seeded the day before transfection to reach optimal confluency, ensuring high viability and maximal transfection efficiency. What are HEK293T cells and why use them? HEK293T cells are a human embryonic kidney-derived cell line engineered to express SV40 large T antigen, which enhances plasmid replication and expression. They are highly transfectable, robust in culture, and support efficient viral particle production - making them the gold standard for viral vector generation. Step 2 - Transfection Plasmid DNA is mixed with PEI (polyethylenimine), a cationic polymer that binds DNA through electrostatic interactions. These PEI-DNA complexes are taken up by cells via endocytosis, allowing the plasmids to enter the cytoplasm and nucleus. Step 3 - Viral production Inside the cells, the plasmids are expressed using the host transcriptional and translational machinery. In lentiviral or retroviral systems, this includes transfer, packaging, and envelope plasmids, leading to assembly and release of viral particles into the culture supernatant. After collecting the virus-containing supernatant, it is centrifuged to remove cell debris. The clarified supernatant can then be used directly (unconcentrated) or further concentrated, depending on the required viral titer and downstream application. The virus is aliquoted into different volumes tailored to experimental needs and stored at −80 °C until use. Step 4 - Cell modification The harvested virus is used to genetically modify target cells. This enables stable gene delivery, such as expression of fluorescent proteins for live-cell imaging or engineering immune cells to express chimeric antigen receptors to generate CAR T cells for cancer immunotherapy. The viral vectors used are engineered for safety and are replication-incompetent, meaning they can deliver genes but cannot make new virus. Key viral genes are split across multiple plasmids and modern vectors are self-inactivating, minimizing risk. #scientist #science #laboratory #immunology #biology
