Replication starts at origins of replication—single in prokaryotes (e.g., OriC in E. coli) and multiple in eukaryotes. Helicase unwinds the DNA, while single-strand binding proteins prevent reannealing. Primase lays down an RNA primer, allowing DNA polymerase to extend new DNA strands in the 5’→3’ direction. The leading strand is synthesized continuously, while the lagging strand forms Okazaki fragments, later joined by DNA ligase after primer removal.
