Here’s a clear, lab-friendly breakdown 👇 (Always follow your lab SOP — steps may slightly vary) 🔬 Step 1: Prepare your cells • Adherent cells → Detach properly, make a single-cell suspension, then centrifuge • Suspension cells → Directly centrifuge • Resuspend the pellet in 0.5–1 mL fresh culture medium 🧼 Step 2: Prep the hemocytometer • Clean and dry the hemocytometer and coverslip properly • Place the coverslip correctly to form the counting chamber 🧪 Step 3: Staining • Mix 10 µL cell suspension + 10 µL Trypan Blue • This gives a dilution factor of 2 🧬 Step 4: Load & focus • Load 10 µL of the mixture into the chamber • Focus first at low magnification, then zoom in 🔢 Step 5: Counting • Count cells in the four corner squares • Count only live cells (clear/unstained) • Dead cells appear blue 📐 Formula to remember Cells/mL = Average cell count × Dilution factor × 10⁴ • Dilution factor here = 2 📝 Why this matters ✔️ Accurate cell seeding ✔️ Assess viability of newly thawed cultures ✔️ Maintain healthy, reproducible experiments 📌 Mastering cell counting = mastering basic cell culture skills.
